To the Editor.—Researchers at Duke University have shown that apolipoprotein E4 (apo E) gene dosage correlates with increased risk of Alzheimer's disease

ApoE is synthesized by the liver and is also made locally in the brain. There are three common variants of apoE, resulting from common genetic variation, called E2, E3 and E4. Apolipoprotein E (APOE) is a protein involved in the metabolism of fats in the body of mammals. A subtype is implicated in Alzheimer's disease and cardiovascular disease. APOE belongs to a family of fat-binding proteins called apolipoproteins. Apolipoprotein E4 is a member of the apolipoprotein E family of plasma lipoproteins that bind reversibly with lipoprotein and play an important role in lipid metabolism. ApoE4 regulates plasma lipid levels by increasing the degradation of particles rich in triglycerides and cholesterol.

Apolipoprotein E enhances proteolytic break-down of this peptide, both within and between cells. The isoform APOE-ε4 is not as effective as the others at promoting these reactions, resulting in increased vulnerability to AD in individuals with that gene variation. A major and the second most common isoform of apolipoprotein E. In humans, Apo E4 differs from APOLIPOPROTEIN E3 at only one residue 112 (cysteine is replaced by arginine), and exhibits a lower resistance to denaturation and greater propensity to form folded intermediates. Apo E4 is a risk factor for ALZHEIMER DISEASE and CARDIOVASCULAR DISEASES. As a member of the wwPDB, the RCSB PDB curates and annotates PDB data according to agreed upon standards. The RCSB PDB also provides a variety of tools and resources.

2019-06-11 · Apolipoprotein (apo) E4, an isoform of the APOE gene in humans, is the major genetic risk factor for late-onset familial and sporadic AD [ 4, 5, 6, 7, 8 ], which account for most AD cases. ApoE4 increases the risk and decreases the age of onset of AD in a gene dose dependent manner [ 4, 5, 6, 7, 8, 9, 10, 11 ].

Apolipoprotein (apo) E plays a fundamental role in the maintenance and repair of neurons, but its three isoforms differ in their abilities to accomplish these critical tasks (1 – 3). ApoE4 is associated with a wide variety of neuropathological processes.

Many studies have demonstrated the detrimental effects of apoE4 in varying cellular contexts. However, the underlying mechanisms explaining how apoE4 leads to cognitive decline are not fully understood.

Here, we suggest that meningeal lymphatic-vessel function is influenced by apolipoprotein E4 (APOE4), which has been established as the leading genetic risk factor for developing AD . We also present novel analyses of previously published RNA-Seq data that offer new insights into how meningeal lymphatic vessels, in association with APOE4, may contribute to the pathogenesis of AD. 2019-03-20 · This calls for a shift in paradigm where targeted treatment is developed for specific AD subpopulations that share distinct genetic or pathological properties. Apolipoprotein E4 (apoE4), the most prevalent genetic risk factor of AD, is expressed in more than half of AD patients and is thus an important possible AD therapeutic target.
